fuel pressure setting

i am running a port injection system on my bike.
i have read somewhere that you set the pressure with the engine off and vacumm pipe connected.
my injectors are set at 43psi. so what should the running pressure be ?
higher than 43 or exactly 43 ?

Less when the engine is under vacuum.
